I would guess perhaps a hotspot or a something like an infection surrounding a splinter etc or fungal infection

Did you switch foods recently? Add a new cleaning product? Perhaps an allergic reaciton to something that she's been rubbing/scratching at

Time to have the vet take a look at it.
